RESUMO

MicroRNAs (miRNAs) play a significant role in axon regeneration following spinal cord injury. However, the functions of numerous miRNAs in axon regeneration within the central nervous system (CNS) remain largely unexplored. Here, we elucidate the positive role of miR-2184 in axon regeneration within zebrafish Mauthner cells (M-cells). The upregulation of miR-2184 in the single M-cells facilitates axon regeneration, while the specific sponge-induced silencing of miR-2184 leads to impeded axon regeneration. We show that syt3, a downstream target of miR-2184, negatively regulates axon regeneration, and the regeneration suppression by syt3 depends on its binding to Ca2+. Furthermore, pharmacological stimulation of the cAMP/PKA pathway suggests that changes in the readily releasable pool may affect axon regeneration. Our data indicate that miR-2184 promotes axon regeneration of M-cells within the CNS by modulating the downstream target syt3, providing valuable insights into potential therapeutic strategies.

We present the performances of a broadband optical parametric chirped pulse amplification (OPCPA) using partially deuterated potassium dihydrogen phosphate (DKDP) crystals with deuteration levels of 70% and 98%. When pumped by a Nd:glass double frequency laser, the OPCPA system using the 98% deuterated DKDP crystal achieves a broad bandwidth of 189 nm (full width at 1/e2 maximum) from 836 nm to 1025 nm. For the DKDP crystal with length of 43 mm, the pump-to-signal conversion efficiency reaches 28.4% and the compressed pulse duration is 13.7 fs. For a 70% deuterated DKDP crystal with a length of 30 mm, the amplified spectrum ranges from 846-1021 nm, the compressed pulse duration is 15.7 fs, and the conversion efficiency is 25.5%. These results demonstrate the potential of DKDP crystals with higher deuteration as promising nonlinear crystals for use as final amplifiers in 100 Petawatt (PW) laser systems, supporting compression pulse duration shorter than 15 fs.

Cooperation between primary malignant cells and stromal cells can mediate the establishment of lung metastatic niches. Here, we characterized the landscape of cell populations in the tumor microenvironment in treatment-naïve osteosarcoma using single-cell RNA sequencing and identified a stem cell-like cluster with tumor cell-initiating properties and prometastatic traits. CXCL14 was specifically enriched in the stem cell-like cluster and was also significantly upregulated in lung metastases compared with primary tumors. CXCL14 induced stromal reprogramming and evoked a malignant phenotype in fibroblasts to form a supportive lung metastatic niche. Binding of CXCL14 to heterodimeric integrin α11ß1 on fibroblasts activated actomyosin contractility and matrix remodeling properties. CXCL14-stimulated fibroblasts produced TGFß and increased osteosarcoma invasion and migration. mAbs targeting the CXCL14-integrin α11ß1 axis inhibited fibroblast TGFß production, enhanced CD8+ T cell-mediated antitumor immunity, and suppressed osteosarcoma lung metastasis. Taken together, these findings identify cross-talk between osteosarcoma cells and fibroblasts that promotes metastasis and demonstrate that targeting the CXCL14-integrin α11ß1 axis is a potential strategy to inhibit osteosarcoma lung metastasis. SIGNIFICANCE: Cooperation between stem-like osteosarcoma cells and fibroblasts mediated by a CXCL14-integrin α11ß1 axis creates a tumor-supportive lung metastatic niche and represents a therapeutic target to suppress osteosarcoma metastasis.

PURPOSE: The purpose of the study was to evaluate the efficacy and safety of tirofiban use in endovascular thrombectomy for intravenous thrombolysis applicable patients of large vessel occlusion stroke with data from Direct-MT trial. MATERIALS AND METHODS: Direct-MT was the first randomized controlled trial to prove the non-inferiority of thrombectomy alone to bridging therapy (intravenous thrombolysis before thrombectomy) for large vessel occlusion stroke. Patients who underwent endovascular procedure were included and divided into thrombectomy-alone group and bridging therapy group. The effect of tirofiban use on 90 days MRS distribution, MRS 0-2 and mortality, successful reperfusion, the ASPECTS and outcome lesion volume of index stroke, re-occlusion of the treated vessel, futile recanalization and safety outcomes were further evaluated in both groups after adjustment for relevant confounding factors. The interaction between tirofiban and rt-PA was also assessed. RESULTS: Of 639 patients included in this analysis, 180 patients underwent thrombectomy with tirofiban use (28.2%). Patients with tirofiban use had lower percentage of bridging therapy (41.1% vs 54.3%, P = 0.003), higher proportion of large artery atherosclerosis (P < 0.001) and more emergent stenting (30.56% vs 6.97%, P < 0.001). After adjustment for confounding factors, the 90-day modified Rankin Scale distribution, successful final recanalization rate, outcome lesion volume of index stroke on CT and intracranial hemorrhage risk showed no difference after tirofiban use in thrombectomy-alone group and in bridging therapy group. No interaction effect between tirofiban and rt-PA was detected. CONCLUSION: Based on data from Direct-MT trial, tirofiban is a safe medication for intravenous thrombolysis applicable patients with large vessel occlusion stroke undergoing thrombectomy. Pre-pulses caused by the post-pulses in the optical parametric chirped-pulse amplifier were comprehensively studied for the first time, including the underlying mechanism for the delay-shift of pre-pulses, the intensity variation of pre-pulses affected by the initial delay of post-pulses and the pump energy, and also the nonlinear beat noise. The simulation and measurement confirmed that the high-order dispersion of the pulse stretcher was the main cause for the delay-shift of pre-pulses, which should be similar with the chirped-pulse amplifiers. The intensity of pre-pulses would decrease significantly as the initial delay of post-pulses increased, but would increase with the growth of pump energy. Moreover, the temporal position of the nonlinear beat noise in the experiment was successfully predicted by our simulation. This work could help us better understand the pre-pulses in OPCPA and provide helpful guidance for designing high-contrast laser systems.

Reactive oxygen species (ROS) are closely related to the antiviral immune response of plants, while virus can regulate ROS through various pathways to facilitate their own infection or replication. Citrus yellow vein clearing virus (CYVCV) is one of the most devastating viruses affecting lemon (Citrus limon) industry worldwide. However, the pathogenesis of CYVCV remains poorly understood. In this study, direct interaction between the coat protein (CP) of CYVCV and the ascorbate peroxidase 1 of lemon (ClAPX1) was confirmed for the first time by yeast two-hybrid, Bimolecular Fluorescence Complementation, and Co-immunoprecipitation assays. Transient expression of CP in lemon and Nicotiana benthamiana significantly enhanced the enzyme activity of the ClAPX1, and then inhibited the accumulation of H2O2. In addition, overexpression of ClAPX1 in lemon by transgene significantly promoted CYVCV accumulation and depressed the expression of most genes involved in jasmonic acid (JA) signaling pathway. Correspondingly, ClAPX1 silencing by RNA interference inhibited CYVCV accumulation and increased the expression of most genes involved in JA signaling pathway. To our knowledge, this is the first report that viruses regulate ROS by targeting APX directly, thereby suppressing host immune response and promoting viral accumulation, which may be mediated by JA signaling pathway.

The temporal contrast of high-peak-power lasers is usually limited by pre-pulses, which are generally produced by post-pulses due to the nonlinearity of the active medium. The reason for the conversion between pre-pulse and post-pulse is now well known, but the mechanisms for the delay-shift and asymmetric broadening of the newly generated pre-pulse are not yet clear. In this work, a novel, to the best of our knowledge, numerical model combining the nonlinear Schrödinger equation and the Frantz-Nodvik equation is proposed to investigate the underlying mechanisms for the "distortion" of the pre-pulse. Numerical results show that the gain characteristics of Ti:sapphire amplifiers can only make a minor change on the temporal profile of the pre-pulse, but the high-order dispersion is the main cause for the delay-shift and asymmetric broadening of the pre-pulse, and the effects are more significant for the initial post-pulse with a relatively larger delay.

Studies on runoff are crucial for the scientific allocation, utilization, and development of water resources and can provide an important basis for the prevention and control of flood and drought disaster, as well as water environmental pollution management. Affected by global warming, the frequency and intensity of extreme climate events, particularly extreme precipitation, have significantly changed in recent years, which can directly or indirectly impact runoff changes. In this study, we used the SWAT model to simulate the spatiotemporal variations in runoff in the Yangtze River Basin from 1965 to 2019 and analyzed the response of runoff to precipitation under extreme conditions. The results showed that the changes in total runoff in the Yangtze River Basin were not significantly different from 1965 to 2019. The total runoff and the mid-lower runoff in the basin experienced four stages of "dry-wet-dry-wet." Simulations revealed that under the 50-year extreme precipitation event, the increase in daily average runoff was 6200%, 21%, and 15% for the typical sub-basins of the upper, middle, and lower reaches of the Yangtze River, respectively. Additionally, the increase in monthly and annual average runoff was 355%, 5%, and 1.3% and 78%, 1%, and 0.24%, for upper, middle, and lower reaches of the Yangtze River, respectively. Moreover, under the 100-year extreme precipitation, the average daily runoff increasing rates were 8000%, 25%, and 17% for upper, middle, and lower reaches of the Yangtze River, respectively, compared to the monthly increase of 437%, 7%, and 1.5% and annual increase of 96%, 1.2%, and 0.28%, respectively. Our findings may improve the understanding of hydrological responses to climate change and provide valuable inferences to decision-makers and water managers for better allocation and management of water resources.

Accurate identification of lesions and their use across different medical institutions are the foundation and key to the clinical application of automatic diabetic retinopathy (DR) detection. Existing detection or segmentation methods can achieve acceptable results in DR lesion identification, but they strongly rely on a large number of fine-grained annotations that are not easily accessible and suffer severe performance degradation in the cross-domain application. In this paper, we propose a cross-domain weakly supervised DR lesion identification method using only easily accessible coarse-grained lesion attribute labels. We first propose the novel lesion-patch multiple instance learning method (LpMIL), which leverages the lesion attribute label for patch-level supervision to complete weakly supervised lesion identification. Then, we design a semantic constraint adaptation method (LpSCA) that improves the lesion identification performance of our model in different domains with semantic constraint loss. Finally, we perform secondary annotation on the open-source dataset EyePACS, to obtain the largest fine-grained annotated dataset EyePACS-pixel, and validate the performance of our model on it. Extensive experimental results on the public dataset FGADR and our EyePACS-pixel demonstrate that compared with the existing detection and segmentation methods, the proposed method can identify lesions accurately and comprehensively, and obtain competitive results using only coarse-grained annotations.

Automatic medical report generation based on deep learning can improve the efficiency of diagnosis and reduce costs. Although several automatic report generation algorithms have been proposed, there are still two main challenges in generating more detailed and accurate diagnostic reports: using multi-view images reasonably and integrating visual and semantic features of key lesions effectively. To overcome these challenges, we propose a novel automatic report generation approach. We first propose the Cross-View Attention Module to process and strengthen the multi-perspective features of medical images, using mean square error loss to unify the learning effect of fusing single-view and multi-view images. Then, we design the module Medical Visual-Semantic Long Short Term Memorys to integrate and record the visual and semantic temporal information of each diagnostic sentence, which enhances the multi-modal features to generate more accurate diagnostic sentences. Applied to the open-source Indiana University X-ray dataset, our model achieved an average improvement of 0.8% over the state-of-the-art (SOTA) model on six evaluation metrics. This demonstrates that our model is capable of generating more detailed and accurate diagnostic reports.

Low-dispersion mirrors (LDMs), which require a broad bandwidth, low dispersion, and high damage threshold, are essential optics in ultra-intense and ultra-short laser devices. Bragg mirrors and chirped LDMs do not satisfy these requirements simultaneously. We propose a novel LDM (NLDM) based on the hump-like structure and quarter wavelength optical thickness (QWOT) structure to achieve a broad bandwidth, smooth dispersion, and high robustness. The spectral and dispersion characteristics of the two structures compensate for each other, which makes up for the deficiency that the dispersion bandwidth of the sinusoidal modulation structure cannot be broadened. Based on this structure, the LDM can achieve a design bandwidth of 240 nm and support the transmission of sub-11-fs pulses. The accuracy of the NLDM is experimentally evaluated. The structure shows the potential for broad-spectrum laser damage performance due to the low electric field intensity. The NLDM improves the mirror performance and paves the way for a new generation of ultra-intense and ultra-short laser devices.

Apoptosis, with a hallmark of upregulated protease Caspase-3, has been frequently imaged with various probes to reveal the therapeutic efficiencies of different drugs. However, activatable molecular probes with programmable self-assembling behaviors that enable enhanced T1-weighted magnetic resonance imaging (MRI) of apoptosis remain scarce. Herein, taking advantage of a CBT-Cys click reaction, we rationally designed a Caspase-3-activatable self-assembling probe Ac-Asp-Glu-Val-Asp-Cys(StBu)-Lys(DOTA(Gd))-CBT (DEVDCS-Gd-CBT) for apoptosis imaging in vivo. After Caspase-3 cleavage in apoptotic cells, DEVDCS-Gd-CBT underwent CBT-Cys click reaction to form a cyclic dimer, which self-assembled into Gd nanoparticles. With this probe, enhanced T1-weighted MR images of apoptosis were achieved at low magnetic fields in vitro, in cis-dichlorodiamineplatinum-induced apoptotic cells and in tail-amputation-simulated apoptotic zebrafish. We anticipate that the smart probe DEVDCS-Gd-CBT could be applied for T1-weighted MRI of apoptosis-related diseases in the clinic in the future.

We demonstrate the stable and flexible light delivery of multi-microjoule, sub-200-fs pulses over a ∼10-m-long vacuumized anti-resonant hollow-core fiber (AR-HCF), which was successfully used for high-performance pulse synchronization. Compared with the pulse train launched into the AR-HCF, the transmitted pulse train out of the fiber exhibits excellent stabilities in pulse power and spectrum, with pointing stability largely improved. The walk-off between the fiber-delivery and the other free-space-propagation pulse trains, in an open loop, was measured to be <6 fs root mean square (rms) over 90 minutes, corresponding to a relative optical-path variation of <2 × 10-7. This walk-off can be further suppressed to ∼2 fs rms simply by using an active control loop, highlighting the great application potentials of this AR-HCF setup in large-scale laser and accelerator facilities.

Shallow groundwater nitrate nitrogen (NO3--N) concentrations in agricultural areas usually show high spatial and intra-annual variability. It is hard to predict such concentrations due to the complexity of influencing factors (e.g., different forms of N in soil, vadose zone characteristics, and groundwater physiochemical conditions). Here, a large number of groundwater and soil samples were collected monthly over two years at 14 sites to analyze the soil and groundwater physiochemical properties and the stable isotopes of δ15N and δ18O of groundwater NO3--N in agricultural areas. Based on field observations, a random forest (RF) model was used to predict the groundwater NO3--N concentrations and reveal the importance of effect factors. The results show that there are large spatiotemporal variations in NO3--N, δ15N-NO3-, and δ18O-NO3- in groundwater. NO3--N is the major dominant specie of inorganic N in groundwater, and the groundwater NO3--N concentration in 24 % of the samples failed to meet the drinking water standard of the WHO (10 mg L-1). The RF model satisfactorily predicted groundwater NO3--N concentrations with R2 of 0.90-0.94, RMSE of 4.54-5.07, and MAE of 2.17-3.38. Groundwater nitrite and ammonium are the most important factors related to NO3--N consumption and production, respectively, in groundwater. Denitrification and nitrification were further identified by the relationships among δ15N-NO3-, δ18O-NO3-, and NO3--N, and by the ranges of δ15N-NO3-, δ18O-NO3-, temperature, pH, DO, and ORP in groundwater. Soil-soluble organic nitrogen (S-SON) and the depth of groundwater table were identified as vital factors related to N sourcing and leaching. Overall, as a first approach to adopting a RF model for high spatiotemporal-resolution prediction of groundwater NO3--N variations, the findings of this study enable a better understanding of groundwater N pollution in agricultural areas. Optimizing management of irrigation and N inputs is anticipated to reduce S-SON accumulation and mitigate the threat to groundwater quality in agricultural areas.

Neural tube defects (NTDs) are severe congenital malformations that can lead to lifelong disability. Wuzi Yanzong Pill (WYP) is an herbal formula of traditional Chinese medicine (TCM) that has been shown to have a protective effect against NTDs in a rodent model induced by all-trans retinoic acid (atRA), but the mechanism remains unclear. In this study, the neuroprotective effect and mechanism of WYP on NTDs were investigated in vivo using an atRA-induced mouse model and in vitro using cell injury model induced by atRA in Chinese hamster ovary (CHO) cells and Chinese hamster dihydrofolate reductase-deficient (CHO/dhFr) cells. Our findings suggest that WYP has an excellent preventive effect on atRA-induced NTDs in mouse embryos, which may be related to the activation of the PI3K/Akt signaling pathway, improved embryonic antioxidant capacity, and anti-apoptotic effects, and this effect is not dependent on folic acid (FA). Our results demonstrated that WYP significantly reduced the incidence of NTDs induced by atRA; increased the activity of catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and content of glutathione (GSH); decreased the apoptosis of neural tube cells; up-regulated the expression of phosphatidylinositol 3 kinase (PI3K), phospho protein kinase B (p-Akt), nuclear factor erythroid-2 related factor (Nrf2), and b-cell lymphoma-2 (Bcl-2); and down-regulated the expression of bcl-2-associated X protein (Bax). Our in vitro studies suggested that the preventive effect of WYP on atRA-treated NTDs was independent of FA, which might be attributed to the herbal ingredients of WYP. The results suggest that WYP had an excellent prevention effect on atRA-induced NTDs mouse embryos, which may be independent of FA but related to the activation of the PI3K/Akt signaling pathway and improvement of embryonic antioxidant capacity and anti-apoptosis.

ETHNOPHARMACOLOGICAL RELEVANCE: Wuzi Yanzong Pill (WYP) is a classic traditional Chinese medicine (TCM) formula that is used for reproductive system diseases. Previous studies showed that WYP had a preventive effect on the development of neural tube defects (NTDs) induced by all-trans retinoic acid (atRA) in mice. AIM OF THE STUDY: This study aimed to determine the optimal combination of main monomer components in WYP on preventing NTDs and to understand the underlying mechanism. MATERIALS AND METHODS: An optimal combination was made from five representative components in WYP including hyperoside, acteoside, schizandrol A, kaempferide and ellagic acid by orthogonal design method. In a mouse model of NTDs induced by intraperitoneal injection of atRA, pathological changes of neural tube tissues were observed by Hematoxylin & Eosin (HE) staining, neural tube epithelial cells apoptosis was detected by terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling (TUNEL), protein changes related to apoptosis, anti-apoptosis, and antioxidant factors were detected with Western blot. Potential targets and mechanisms of monomer compatibility group (MCG) acting on NTDs were analyzed by bioinformatics. RESULTS: Treatment with different combinations of WYP bioactive ingredients resulted in varying decreases in the incidence of NTDs in mice embryos. The combination of MCG15 (200 mg/kg of hyperoside, 100 mg/kg of acteoside, 10 mg/kg of schizandrol A, 100 mg/kg of kaempferide and 1 mg/kg of ellagic acid) showed the most significant reduction in NTD incidence. Mechanistically, MCG15 inhibited apoptosis and oxidative stress, as evidenced by reduced TUNEL-positive cells, downregulation of caspase-9, cleaved caspase-3, Bad, and Bax, and upregulation of Bcl-2, as well as decreased MDA and increased SOD, CAT, GSH, HO-1, and GPX1 levels. Bioinformatics analysis showed that MCG15 acted on the PI3K/Akt signaling pathway, which was confirmed by Western blot analysis showing increased expression of p-PI3K, p-Akt/Akt, and Nrf2 related indicators. CONCLUSION: We have identified an optimal combination of five bioactive components in WYP (MCG15) that prevented NTDs in mice embryos induced by atRA by activating the PI3K/Akt signaling pathway and inhibiting apoptosis and oxidative stress.

OBJECTIVES: To review the pharmacokinetics, pharmacological action and mechanism of isoalantolactone (IAL). Explore the therapeutic potential of isoalantolactone.Keywords including isoalantolactone, pharmacological effects, pharmacokinetic and toxicity were used for literature search in PubMed, Excerpta Medica Database (EMBASE) and Web of Science, to identify articles published from 1992 to 2022. KEY FINDINGS: IAL has a great many obiological activities such as anti-inflammatory, antioxidant, antitumour, neuroprotection, with no obvious toxicity. This review suggests that IAL exerts different pharmacological effects with different mechanisms of action at different doses, and may be a potential drug candidate to treat inflammatory diseases, neurodegenerative diseases and cancer, with medicinal value. SUMMARY: IAL has various pharmacological activities and medicinal values. However, further research is needed to determine its specific intracellular action sites and targets, so as to fully understand its therapeutic mechanism and provide a reference for the treatment of related diseases.

Antarctic krill (Euphausia superba) is Earth's most abundant wild animal, and its enormous biomass is vital to the Southern Ocean ecosystem. Here, we report a 48.01-Gb chromosome-level Antarctic krill genome, whose large genome size appears to have resulted from inter-genic transposable element expansions. Our assembly reveals the molecular architecture of the Antarctic krill circadian clock and uncovers expanded gene families associated with molting and energy metabolism, providing insights into adaptations to the cold and highly seasonal Antarctic environment. Population-level genome re-sequencing from four geographical sites around the Antarctic continent reveals no clear population structure but highlights natural selection associated with environmental variables. An apparent drastic reduction in krill population size 10 mya and a subsequent rebound 100 thousand years ago coincides with climate change events. Our findings uncover the genomic basis of Antarctic krill adaptations to the Southern Ocean and provide valuable resources for future Antarctic research.

Common prosperity is an important feature of Chinese-style modernization. The difficulty and focus of promoting the construction of common prosperity in China lies in rural areas and rural households. How to evaluate the common prosperity of rural households is becoming an important research topic. Based on the perspective of meeting the people's needs for a better life, this study designed 14 items or indicators from the dimensions of affluence, commonality, and sustainability. The common prosperity of rural households is regarded as a potential structure. Based on the survey data of 615 rural households in Zhejiang Province, graded response models were used to estimate the discrimination and difficulty coefficient, and an indicator selection and characteristics analysis were carried out. The research results show that there are 13 items suitable for measuring the common prosperity of rural households, and these indicators have strong distinguishing ability. However, different dimension indicators have different functions. In particular, the affluence dimension, the sharing dimension, and the sustainability dimension are suitable for distinguishing families with a relatively high level of common prosperity, a medium level of common prosperity, and a low level of common prosperity, respectively. Based on this, we propose policy recommendations such as building diversified governance strategies, formulating differentiated governance policies, and supporting the corresponding basic policy reform.

Saline-alkali stress is a universal abiotic stress factor limiting fruit tree cultivation worldwide. Apple (Malus×domestica Borkh.) is one of the fruits with the largest yields worldwide. Tea crabapple (Malus hupehensis Rehd. var. pingyiensis Jiang) is a type of common apple rootstock in China. Because facultative apomixis occurs in this species, it is often used in molecular research. The present study investigated the molecular mechanism of the response of indoleacetic acid (IAA) and cytokinins [zeatin, trans-zeatin riboside (tZR), isopentenyladenine (iP), and isopentenyladenosine (iPA)] to mixed saline-alkali stress (MSAS) in tea crabapple leaves. The endogenous hormone content of tea crabapple leaves under MSAS was measured, and the expression of stress response-related genes was analyzed by RNA sequencing. The results showed that the concentration of IAA was initially higher and then lower than that in the control, whereas the concentration of zeatin, tZR, iP, and iPA was higher than that in the control. A total of 1262 differentially expressed genes were identified in the three comparison groups. Further analyses suggested that IAA and cytokinin biosynthetic genes were mostly upregulated in tea crabapple leaves, indicating that auxin and cytokinin signaling pathway regulation occurred in response to MSAS. These findings suggest that IAA and cytokinins play an important role in the response of tea crabapple to MSAS.
